bioRxiv · A Fibroblast State Choreographs an Epithelial YAP-dependent Regenerative Program Essential to (Pre)malignancy via ECM-mediated MechanotransductionChronic lung injury generates metaplasia which occasionally, but ominously, progresses to squamous dysplasia and squamous lung cancer. To identify mechanisms through which disrupted tissue homeostasis contributes to malignant initiation and progression, we used in vivo and in vitro heterotypic recombinant models of human bronchial epithelial cells (hBECs) and fibroblasts. We demonstrate that injury-associated TGF-β signaling creates a fibroblast state dependent upon HSP47 upregulation. These fibroblasts accumulated collagen, thus elevating tissue stiffness and activating mechanosignaling that sustained YAP-dependent embryonic-like, pro-malignant activities in adjacent hBECs. This Stress/Tension-Instructive Fibroblast (STIF) state, exhibited by stressed fibroblasts in premalignant and malignant lesions across multiple cancer types, was sufficient to reprogram disease-free hBECs to metaplasia and to drive hBECs with compromised tumor suppressor function to dysplasia, yet could be inhibited and reversed. STIFs suffice to activate epithelial phenotypes reminiscent of oncogene-mediated cell transformation and induce (pre)malignancy via increased force transmission, providing novel targets for prevention. ### Competing Interest Statement The authors have declared no competing interest. As a major structural component of the tumor microenvironment, the ECM plays crucial roles in providing mechanical support, modulating the microenvironment, and serving as a reservoir for signaling molecules. Tissue stiffness is primarily determined by the abundance and cross-linking of the ECM components. Collagen is one of the major components of ECM and contributes to 30% of the total ECM. Increased collagen deposition contributes to enhanced tissue stiffness and is associated with a more aggressive cancer phenotype. A restructured ECM, particularly collagen content, can influence cellular signaling pathways through interaction with cell surface receptors. In the current study, we investigated the role of collagen in prostate cancer. Our findings indicate that collagen, a major component of the TME, modulates prostate cancer cell migration, alters sensitivity to chemotherapeutic agents, and promotes non-genomic activation of androgen receptor signaling through FAK activation, thereby contributing to therapy (androgen deprivation therapy) resistance and cancer progression. ### Competing Interest Statement The authors have declared no competing interest.

To the configuration of bassist Michel Benita and drummer Sebastian Rochford (with whom he previously recorded as Trio Libero) he now welcomes the ambient touch of guitarist Eivind Aarset. The latter, perhaps more than any other, evokes the encompassing waters of the album’s title, and draws on the relationship formed on Sheppard’s ECM debut, Movements in Colour.

Emphasis sides with Sheppard’s compositions, which have the first and final word on Surrounded by Sea. “Tipping Point,” co-written with Benita, opens the set on a distant shore.."

Nowhere within these relatively brief tunes will you find demonstrative solos or waving of virtuosic flags. Yellow Fields is an album by German double bassist and composer Eberhard Weber recorded in September 1975 and released on ECM the following year. The quartet features saxophonist Charlie Mariano, pianist Rainer Brüninghaus and drummer Jon Christensen.

The Penguin Guide to Jazz awarded it the maximum four stars and placed it in their Core Collection, writing "Weber's masterpiece is essentially a period piece which nevertheless still seems modern. The sound of it is almost absurdly opulent: bass passages and swimming keyboard textures that reverberate from the speakers, chords that seem to hum with huge overtones. "For its third ECM outing, pianist Carla Bley’s trio with saxophonist Andy Sheppard and bassist Steve Swallow mixes up an antidote for these times of uncertainty and quarantine. The title suite is the first of three comprising the program. Given that “Life Goes On” came out of a recent brush with illness, it’s fitting that Bley should begin in the dark whimsy of the blues. Her left hand plows fertile soil before leaving Sheppard and Swallow to sow their thematic crop. Years of experience and collaboration funnel into Swallow’s intimate rapport with Bley and into Sheppard’s unforced, spiritual playing. The latter, whether breathing through tenor or soprano, takes two steps forward for every retreat.."
